Gold nanospheres pharmacokinetic bound lanreotide octapeptide as a delivery system for diagnosis
Abstract
Gold nanospheres have advantages as phototherapeutic in cancer treatment, transportation and selective vectoring macromolecules drugs and therapeutic agents. In this project we present nanodrugs pharmacokinetics (AuNPs and AuNPs-LAN) using the analytical method reported in vitro of characterization (Molina-Trinidad, et al.) To determine the dosage of these compounds useful in the diagnosis and treatment cancer, we used a nonlinear fitting program (WinNonlin). The AuNPs follow a two compartment open model (MADC) and AuNPs-LAN conforms to an independent continuous infusion model fit. Pharmacokinetic parameters were obtained for AuNPs (ke = 0.020 ± 0.0016 min-1); (AUC0∞ = 1367 ± 107.3 mg / mL* min); (Vd = 69.49 ± 0.113 mL at steady state); (t1/2 = 254 ± 81 min); (CL = 0.119 mL / min ± 0.0098 mL / min) (MRT = 380.7 ± 150 min). For the conjugated AuNPs-LAN; (λ= 0.328 ± 0.072 min-1); (t1/2 = 2.3 ± 0.0269 h); (AUC0∞ = 9720 ± 1330 μg/ mL *min); (Vd = 1.625 ± 0.182 mL); (CL = 0.527 ± 0.083 mL / min); (MRT = 66.91 ± 8.162 min). It was concluded that pharmacokinetic parameters are useful in subsequent studies (preclinical and clinical phases).
