Thermodynamic profile of the metformin-vanadyl system by isothermal titration calorimetry

Abstract

Metformin is a biguanide that has been used for the last 40 years for the treatment of type 2 diabetes mellitus (DM2). It can form coordination complexes with some metal ions, such as the vanadyl ion. In the scientific literature, it is reported that drugs complexed with metal ions have greater biological activity, so the objective of this work was to evaluate the formation of complex between metformin and the vanadyl ion, determining the thermodynamic parameters associated with this process by Isothermal Titration Calorimetry (ITC) at 25 °C. The determined thermodynamic parameters are: the equilibrium constant (β), the Gibbs
free energy (ΔG°), the enthalpy (ΔH°) and the entropy (ΔS°). In addition, the stoichiometry of the complex formed in the system was also determined. The results obtained are: log β=3.520 ± 0.037, ΔG°= −4.810 kcal/mol, ΔH°= (−0.202 ± 0.006) kcal/mol and −TΔS= −4.610 kcal/mol; the stoichiometry obtained was 1:1 corresponding to the Mtf(VO)2+ complex.
